Supplemental Table 1. Drug Response Profiles for Pharmacogenomics Databases. Drug response profiles for a given cell line and drug from each database (CTRP, GDSCv1, GDSCv2, PRISM). Calculated drug response profiles: log_IC50, log_EC50, AUT, AUC, and AreaTruncAUC are reported for both full and truncated dose ranges. Parameters derived from the fitted sigmoidal curve: R2, RMSE, bottom asymptote of viability (Bottom), and Hill Slope (nH) are also included. 

Supplemental Table 2. Database Pairwise Reproducibility Spearman Correlations for All Drugs. For each database pair (database 1|database 2), the number of pairs with exact number of unique cell lines and drugs (n_unique_drugs, m_unique_cells) used to calculate pairwise Spearman (spearman_r) and Pearson (pearson_r) correlations are reported. Correlations are categorized by response metric (IC50, EC50, AUT, and AUC), dose range (Full or Trunc), and area range (Full or Trunc). The upper and lower bounds of 95% confidence intervals of the bootstrapped Spearman (spearman_r_95CI_lower, spearman_r_95CI_upper) and Pearson (pearson_r_95CI_lower, pearson_r_95CI_upper) correlations are also reported.

For each database pair (database 1|database 2), the number of pairs with exact number of unique drugs and cell lines (n_unique_drugs, m_unique_cells) used to calculate pairwise Spearman (spearman_r) and Pearson (pearson_r) correlations are reported. Correlations are categorized by response metric (IC50, EC50, AUT, and AUC), drug grouping by response (below-median per metric or 1nM < IC50 < 100uM), dose range (Full or Trunc), and area range (Full or Trunc). Corresponding differences between drug-group correlations and those calculated from all drugs are included (delta_spearman_r_all_drugs, delta_pearson_r_all_drugs). The upper and lower bounds of 95% confidence intervals of the bootstrapped Spearman (spearman_r_95CI_lower, spearman_r_95CI_upper) and Pearson (pearson_r_95CI_lower, pearson_r_95CI_upper) correlations are also reported.

Supplemental Table 4. PRISM Runs 21 – 26 Drug Response Profiles. PRISM runs 21 – 26 drug response profiles for a given cell line and drug. Calculated drug response profiles: log_IC50, log_EC50, AUT, AUC, and AreaTruncAUC are reported for both full and truncated dose ranges per PRISM run. Parameters derived from the fitted sigmoidal curve: R2, RMSE, bottom asymptote of viability (Bottom), and Hill Slope (nH) are also included.
